چکیده

N uclear factor (NF)-kB is a key player in the control of both innate and adaptive immunity. NF-kB proteins are present in the cytoplasm in association with inhibitory proteins called inhibitors of kB (IkBs). On activation by a large plethora of inducers, the IkB proteins are phosphorylated, ubiquitinated, and subsequently degraded in the proteasomes. Degradation of IkBs allows translocation of NF-kB into the nucleus and bind to their cognate DNA binding sites to regulate the transcription of large numbers of genes including antimicrobial peptides, cytokines, chemokines, stress response proteins, and antiapoptotic proteins. NF-kB activity is essential for lymphocyte survival, activation, and mounting normal immune responses. Constitutive activation of NF-kB pathways is often associated with inflammatory diseases like rheumatoid arthritis, inflammatory bowel disease, multiple sclerosis, and asthma. Better understanding of the regulation of NF-kB will provide a platform for development of specific therapeutic agents targeted towards the inflammatory diseases.

Effects of adenosine A2a receptor agonist and antagonist on cere-bellar nuclear factor-kBexpression preceded by MDMA toxicity

Background :Adenosine is an endogenous purine nucleoside that has a neuromodulatory role in the central nervous system. The amphetamine derivative (±)-3,4-methylenedioxymethamphetamine (MDMA or ecstasy) is a synthetic amphetamine analogue used recreationally to obtain an enhanced affiliated emotional response. MDMA is a potent monoaminergic neurotoxin with the potential of damage to brain neu...

Phosphorylation of NF-kB and IkB proteins: implications in cancer and inflammation

Nuclear factor-kB (NF-kB) is a transcription factor that has crucial roles in inflammation, immunity, cell proliferation and apoptosis. Activation of NF-kB mainly occurs via IkB kinase (IKK)-mediated phosphorylation of inhibitory molecules, including IkBa. Optimal induction of NF-kB target genes also requires phosphorylation of NF-kB proteins, such as p65, within their transactivation domain by...

Potent Anti-Inflammatory Activity of Tetramethylpyrazine Is Mediated through Suppression of NF-k

The purpose of the current study was to evaluate the anti-inflammatory activity of tetramethlpyrazine on oxazolone-induced colitis mice. Spleen mononuclear cells (SMC), lamina propria mononuclear cells (LPMC) and peripheral blood mononuclear cells (PBMC) were isolated from oxazolone-induced colitis and normal mice. The colitis cells treated by oxazolone were randomly divided into model, low dos...

New Insights into the Role of Nuclear Factor-kB, a Ubiquitous Transcription Factor in the Initiation of Diseases

Nuclear factor-kB (NF-kB) is a ubiquitous transcription factor that governs the expression of genes encoding cytokines, chemokines, growth factors, cell adhesion molecules, and some acute phase proteins in health and in various disease states. NF-kB is activated by several agents, including cytokines, oxidant free radicals, inhaled particles, ultraviolet irradiation, and bacterial or viral prod...
